A groundbreaking development in malaria treatment has emerged from the University of Dundee, where researchers have identified a new compound that could revolutionise the management of this deadly disease. Known as cabamiquine, this innovative molecule may significantly simplify treatment protocols, which is critical as malaria continues to pose a severe threat to public health worldwide.
The Current Malaria Landscape
Malaria remains a leading cause of morbidity and mortality, particularly in tropical regions. According to global health statistics from 2024, there were approximately 282 million cases of malaria, resulting in around 610,000 deaths. These alarming figures reflect a growing challenge, especially with the rising resistance to conventional antimalarial medications. The urgent need for novel treatment options has never been more pronounced.
Introducing Cabamiquine
The newly discovered compound, cabamiquine, is hailed as a “novel drug-like molecule” that can potentially be administered in a single dose. Early clinical trials demonstrated that when combined with the established drug pyronaridine, cabamiquine not only proved effective but also exhibited a commendable safety profile among adults and adolescents.
Dr. Beatriz Baragana, a key member of the research team, expressed optimism regarding the compound’s potential. “This is an exciting step forward for a compound which has great potential to simplify dosing regimens for malaria,” she stated. “The initial work that ultimately led to the discovery of cabamiquine was conducted here in Dundee, with crucial support from Medicines for Malaria Venture (MMV).”
A Step Towards Simplified Treatment
The implications of cabamiquine’s single-dose administration are profound. Professor Ian Gilbert, head of the University of Dundee’s drug discovery unit, highlighted that effective delivery of treatment in areas with inconsistent healthcare access is vital. “The benefits of this compound are not only that it looks to be effective, with a good benefit/risk profile, but it can also be delivered in a single dose,” he explained. This is particularly important for regions where logistical challenges hinder regular healthcare services.
Looking ahead, MMV plans to advance the cabamiquine-pyronaridine combination into further studies, aimed at expanding its application to paediatric populations as well. Paul Willis, vice president of drug discovery and preclinical safety at MMV, emphasised the urgency of developing simplified dosing regimens. “Novel treatment and prevention regimens are urgently needed to enable effective case management and support long-term malaria control and elimination goals,” he remarked.
Collaborative Efforts and Future Directions
The research into cabamiquine is part of a broader collaborative effort involving the pharmaceutical firm Merck and the PAMAfrica consortium, which aims to address malaria in Africa. The study was co-funded by the EDCTP2 Programme, with additional backing from the European Union and the Swedish International Development Co-operation Agency. The collaborative nature of this research underscores the global commitment to tackling malaria and improving health outcomes.
