A novel personalised vaccine has demonstrated the ability to significantly reduce the recurrence of high-risk melanoma in patients following surgical intervention, marking a potentially transformative step in cancer therapeutics. Developed collaboratively by Merck & Co (known as MSD in Europe) and Moderna, the vaccine, named Intismeran, was administered alongside the established immunotherapy drug Keytruda in a phase three clinical trial. The trial engaged over 1,000 participants diagnosed with high-risk stage two, three, or four melanoma—ranging from large tumours confined to a specific area to advanced cancers that have metastasised to other sites. As patients received the new vaccine in conjunction with Keytruda, preliminary findings indicated a marked increase in the duration of cancer-free intervals compared to those who were treated with Keytruda alone. Both companies involved in the development are exploring similar personalised vaccine strategies for other cancer types, including lung, bladder, and kidney cancers. Stéphane Bancel, CEO of Moderna, described the findings as a “pivotal moment” in cancer research, with the potential to redefine therapeutic strategies in oncology.

While the early results are promising, the scientific community remains vigilant. Dr Talisia Quallo, head of prevention and early detection at Cancer Research UK, acknowledged the potential of the research but emphasised the necessity for comprehensive data analysis and continued funding for studies that could lead to innovative, personalised treatment options. Dr Lennard Lee, a consultant medical oncologist and associate professor at the University of Oxford, noted that “this is certainly good news for patients with melanoma. The study has much wider implications as it provides proof of principle that personalised cancer vaccines work.”

The forthcoming presentation of more detailed findings at an upcoming medical conference in October is anticipated with keen interest, as it could further elucidate the efficacy and application of this promising treatment.
